The KeyTec® TR-FRET ALK/CRBN PROTAC Binding Assay Kit is designed for screening PROTAC molecules that mediate the formation of a ternary complex between ALK protein and the CRBN/DDB1 complex. This kit utilizes KeyTec® TR-FRET technology in a homogeneous immunoassay format, offering advantages such as simplicity, speed, high accuracy, and excellent reproducibility.
The basic principle of this method is illustrated below: The KeyTec® TR-FRET Solar Eu1-conjugated anti-Tag1 antibody specifically recognizes the Tag1-CRBN/DDB1 protein complex, while the KeyTec® TR-FRET LA2-conjugated anti-Tag2 antibody specifically recognizes Tag2-ALK protein. In the presence of the positive control PROTAC molecule TRI-611, the Tag1-CRBN/DDB1 complex and Tag2-ALK protein are brought into close proximity, forming a ternary complex. This allows Solar Eu (donor) and LA (acceptor) to come near each other, enabling energy resonance transfer upon external light excitation. By measuring the signal intensity at a specific wavelength (665 nm), the PROTAC-mediated binding between ALK and CRBN/DDB1 can be quantitatively determined.
